Two Design Options
We can automatically synthesize PNA/DNA chimeras in any desired sequence online, mainly including 5’-DNA-3’-PNA and H-PNA-5’-DNA-3’.
- 5’-DNA-3’-PNA. Firstly, PNA is synthesized; secondly, specific building blocks are coupled to the amino end of the PNA chain, and finally the DNA part is synthesized.

- H-PNA-5’-DNA-3’. Firstly, DNA is synthesized; secondly, specific building blocks are coupled to the amino end of the DNA chain, and finally the PNA part is synthesized.

Choice of Linker between PNA and DNA
In the design of PNA/DNA chimeras, in order to obtain chimeras with high binding affinity to complementary nucleic acids, a key step is to choose a suitable linker between PNA and DNA. In principle, two types of linker can be considered. The first type of linker does not contain nucleobases, while the second type of linker contains nucleobases and can bind to complementary nucleic acids.
